چکیده

Oxaliplatin (OXA) resistance limits the efficiency of treatment for hepatocellular carcinoma (HCC). Studies have shown that PDZ-binding kinase (PBK) plays important roles in tumors. However, role PBK HCC is still a problem. In this study, we explored whether involved chemoresistance to OXA HCC. Expressions six cell lines and one human hepatocytes line were determined by real-time quantitative PCR western blot analysis. SNU-182 HepG2 cells chosen induce resistance. was silenced or overexpressed OXA-resistant sensitive lines. Then, proliferation, migration, invasion measured cholecystokinin-8 assay Transwell assay, respectively. The Cancer Genome Atlas dataset showed highly expressed signifies poor prognosis patient with Results expression significantly higher than THLE2 cells, it further increased cells. Silencing promoted sensitivity drug-resistant OXA. Overexpression relieved apoptosis induced migration OXA-sensitive Thus, study revealed high correlated which may provide promising therapeutic target treating

ID1 promotes hepatocellular carcinoma proliferation and confers chemoresistance to oxaliplatin by activating pentose phosphate pathway

BACKGROUND Drug resistance is one of the major concerns in the treatment of hepatocellular carcinoma (HCC). The aim of the present study was to determine whether aberrant high expression of the inhibitor of differentiation 1(ID1) confers oxaliplatin-resistance to HCC by activating the pentose phosphate pathway (PPP). METHODS Aberrant high expression of ID1 was detected in two oxaliplatin-resi...

miR-92a promotes hepatocellular carcinoma cells proliferation and invasion by FOXA2 targeting

Objective(s): MicroRNAs (miRNAs) are considered as powerful, post-transcriptional regulators of gene expression in hepatocellular carcinoma cells (HCC). However, the function of miR-92a is still unclear in HCC. Materials and Methods: Expression of miR-92a in human HCC cell lines was evaluated using qRT-PCR. MTT assay and transwell assay were used to examine the function of miR-92a in HepG2 and ...

Ribosomal RACK1 promotes chemoresistance and growth in human hepatocellular carcinoma.

Coordinated translation initiation is coupled with cell cycle progression and cell growth, whereas excessive ribosome biogenesis and translation initiation often lead to tumor transformation and survival. Hepatocellular carcinoma (HCC) is among the most common and aggressive cancers worldwide and generally displays inherently high resistance to chemotherapeutic drugs. We found that RACK1, the r...

CDCA7L promotes hepatocellular carcinoma progression by regulating the cell cycle.

The cell division cycle-associated 7-like protein (CDCA7L) is a recently-identified target gene of c-Myc which can also interact with c-Myc. It is known to be upregulated in many tumors, however, its role in tumor progression remains unclear. We investigated the role of CDCA7L expression in hepatocellular carcinoma (HCC). We confirmed that CDCA7L is strongly upregulated in human HCC, and demons...

Aurora-A promotes chemoresistance in hepatocelluar carcinoma by targeting NF-kappaB/microRNA-21/PTEN signaling pathway

Hepatocellular carcinoma (HCC) is highly resistant to chemotherapy. Previously, we have shown that Aurora-A mRNA is upregulated in HCC cells or tissues and silencing of Aurora-A using small interfering RNA (siRNA) decreases growth and enhances apoptosis in HCC cells. However, the clinical significance of Aurora-A protein expression in HCC and association between Aurora-A expression and HCC chem...
